Neonatal fibroblast affect the stemness of interfollicular epidermal cells by modulation of basement membrane

Abstract
Fetal wounds reepithelialize rapidly but the mechanism is still unknown. In this study, the epidermal regenerative effects of neonatal fibroblast (n-fibroblasts) were compared to that of child fibroblasts (c-fibroblasts) using a skin equivalent model, which was constructed using interfollicular epidermal stem cells and transit amplifying cells, respectively. Results showed that n-fibroblasts increased the layers of epidermis when compared to c-fibroblasts. An increased number of p63 and PCNA positive cells were observed in SA/n-fibroblasts models {skin equivalent constructed with transit amplifying cells (SA cells) and n-fibroblasts} when compared to SA/c-fibroblasts models {skin equivalent constructed with SA cells and c-fibroblasts}. Expression of both ¥â1 integrin and ¥á6 integrin were increased by n-fibroblasts. The differentiation status was not significantly different. These findings suggested that n-fibroblasts can increase the stemness and proliferative potential of epidermal cells and n-fibroblast can be a source for cell therapy especially for chronic wounds.
